Background

When multiple drugs are taken at the same time, unexpected drug-drug interaction (DDI) may occur between the drugs. The effects of DDI on treatment can be harmful or beneficial. Harmful DDI can reduce the drug efficacy, cause serious side effects, and even increase toxicity; on the other hand, beneficial DDI can enhance efficacy, synergistic drug combinations have been successfully applied in the treatment of complex diseases such as cancer and AIDS.

At present, only a small amount of DDIs were identified in the experimental stage, and most DDIs were discovered through clinical data feedback from patients after the drug was on the market. Due to the serious consequences of improper combination medication, such as the occurrence of medical accidents and the delisting of certain drugs (mibefradil, astemizole, etc.), combination medication has attracted more and more attention, the need for research on DDI before listing is becoming urgent, which is of great significance for improving the safety of clinical combination drugs, the development of new drugs, and the improvement of people's quality of life.
