Reverse Mammalian Protein-Protein Interaction Trap (Reverse MAPPIT) Service

Introduction

Proteins are the executors of most life activities, and the interaction between proteins is the core of cell mechanisms. Therefore, changes in the interaction caused by abnormal protein expression patterns or mutations will trigger cell dysfunction and ultimately lead to diseases. In addition, many viral and bacterial pathogens rely on protein-protein interactions to exert their destructive effects.

The Reverse Mammalian Protein-Protein Interaction Trap (Reverse MAPPIT) is a mammalian reverse two-hybrid technology. This method is a two-hybrid complementary system modified by the forward MAPPIT technology, in which a positive readout is generated when the protein-protein interaction is interrupted. The interaction between bait and inhibitory prey (i-prey) blocks signaling and reporter induction. Only through competing proteins or small organic molecules disrupting their interactions can the signaling be restored, leading to a positive readout. With the help of an effective screening platform, this method can be applied to screen for interferences of specified protein-protein interactions in intact mammalian cells. In human cells, it can analyze the destruction, toxicity, and permeability of the test compounds, and is suitable for screening protein-protein interactions of interest in targeted therapy or mapping interaction interface between proteins.
